Abstract
Agro-forestry system combines forestry and agronomy to create a harmony between agricultural intensification and environment preservation. It is because it contains agricultural plants having commercial values, such as spices and coffee, and also creates an opportunity for other plants. Such as: coffee trees alternated with flowering trees of Erythrina spp, which has function as a shade of coffee trees and also to satisfy farmer wood needs. It is not differ from the agro-forestry form above, this research uses a simple agro-forestry form using oranges tree alternated with seasonal plants that is soybean. Soybean plant is classified into sub-family of Papilionoideae with root-nodules. Bradyrhizobium japonicum utilization represents an attempt to add Rhizobium bacteria into the land that appropriate for soybean plant in order to be able to tie up N2 maximally from the air to satisfy N needs of plant and then it can increase soybean result. The purpose of this research is to isolate, to select, and to describe Bradyrhizobium japonicum under an upright cluster of orange plants. This research is conducted in Sengguruh Village, sub-district of Kepanjen, Malang Regency, and in biotechnology laboratory of Muhammadiyah University of Malang since April until August 2007. Tools used on the land include mattock, sickle, gauge, raffia, paper, ballpoint, plastic, and a tool to make a hole on land (gejik). Tools used in the laboratory are LAFC (Laminar Air Flow Cabinet), bunsen, match, autoclave, incubator, analytical weights, gas stove, microwave, petridis, tweezers, Ose needles, beaker glass, Erlenmeyer, and culture bottles. Materials used on the land include variety of Wilis, Anjasmoro, Kaba, Burangrang, and Sinabung soybean seeds. Materials used on the laboratory are nodule, 96% ethanol, 70% alcohol, 0.1% MgCl2, Aquadest, sterile aquadest, agar, Pepton, Paper, Plastic and MgSO4. The performed method in this research is planting soybean with 5 different varieties under the shade or an upright of cluster of orange plants. Inoculation process was conducted in Biotech laboratory of UMM after the nodule of soybean plant under orange plants upright of cluster is taken randomly. The research result shows that it has been obtained 5 superior strains of Bradyrhizobium japonicum. Their best growth rate is on Agar Pepton media with storage temperature in incubator is 28°C for 1 week planting. From this research, the character of Bradyrhizobium japonicum is also obtained. This plant is able to make mutualism symbiosis with soybean plant, has negative gram quality and has bar form, growth quickly. Colonies’ color is varied start from white to yellowish white. Colonies’ thickness is also varied, start from thin to very thick. Colony species and top of colony are also differ, that is colony with little mucous to a lot of mucous, and so do colonies’ top that is dome-shaped, branched, a lot of branches.
