Abstract

ABSTRACT Juliana, Triaini Fadjrin. 2019. Potential Effectiveness of Nut-Grass Root Extract (Cyperus rotundus L) on Increasing Early Apoptotic Cell Line HL-60 in Acute Myeloblastic Leukemia. Thesis. Faculty of Medicine, University of Muhammadiyah Malang. Advisors: (1) dr. Desy Andari, M.Biomed (2) Dr.dr.Fathiyah Safithri, M.Kes Background: Leukemia, especially the AML type, was one of the non-communicable diseases with a high rate of deaths which mollecularly has a defect on cell cycle, especially in the apoptosis pathway. The content of apigenin, luteolin, and quercentin in the nut-grass root extract can induce pro-caspase activation thereby increased the incidence of early apoptotic in AML-type leukemic cells. Objective: To determine the potential effectiveness of Nut-Grass root Extract (Cyperus rotundus L) on cell line HL-60 as a trigger for early apoptosis in Acute Myeloblastic Leukemia. Method: Experimental in vitro using The Post Test Only Control Group Design. The sample of the study used the cell line HL-60 which were divided into 5 groups. Data analysis using One Way ANOVA, Post Hoc Bonferroni and linear regression test. Research Results and Discussion: The doses of Nut-Grass root extract were 38.4 µg / mL, 384 µg / mL, and 3840 µg / mL have been proven Nut-Grass root extract were significant on increased the number of early apoptotic process (sig = 0,000, p <0,05). Dose of 384 µg / mL began to shown significant increased (sig = 0,000). The effect of Nut-Grass root extract were increased the early apoptotic process by 52% (R2 = 0,520). Apigenin, luteolin, and quercentin compounds were capable to activated caspase 8 resulted in early apoptotic which was showed by reversed the phosphatydilserine protein membrane cell. Conclusion: Nut-Grass Root Extract (Cyperus rotundus L) can increase early apoptosis in Leukemia HL-60 cell line Acute Myeloblastic Leukemia.
