Abstract

The results of Horticultural statistical data show that yields are declining and land area is getting tighter. Meanwhile the number of basic agricultural needs increases with the increase in population. Potatoes (Solanum tuberosum L.) is one of the important commodities to be developed. Development of potato varieties needs to be done to get superior varieties that are free of pathogens, can be accepted and produced in the required planting land on dry land. In vitro culture techniques have now developed into a very important plant technique in various plant species. The use of mannitol as an osmoregulator for potatoes can be done in vitro which is able to get a culture that is free of viruses. This study aims to produce potato varieties that can withstand drought stress conducted from February 2019 to June 2019 at the In Vitro Structure Labolatorium (Biotechnology), University of Muhammadiyah Malang. The research method was carried out using a Randomized Group Design that was repeated 3 times. The First Factor consists of 2 levels, namelyv V1 = Lembang Granola Varieties and V2 = Kembang Granola Varieties. The second factor is the media with mannitol concentration, which consists of 6 levels namely K0 = 0 g / l, K1 = 20 g / l, K2 = 40 g / l, K3 =, 60 g / l, K4 = 80 g / l, K5 = 100 g / l. Based on the results of research carried out obtained potato planlet at each concentration of mannitol is different that can affect the overall parameters in this study, among other times appear shoots, plant height, number of roots, number of leaves, number of segments, chlorophyll, dry weight, dry weight , stomata and plant vigor.
