Abstract

Potatoes (Solanum tuberosum L.) is one of the important vegetable plants in Indonesia that has long been known to the public. By promoting a food diversification campaign and developing a snack food industry that uses potatoes as staples, it increasingly makes this plant an important horticultural commodity. Potato cultivation must be considered in certain areas. All related to potatoes that are tolerant of stress. Treatment PEG in explants aims to create drought stress conditions in the environment around explants. The purpose of this study was to determine the differences in growth characteristics of potato explants against drought stress conditions and provide information about PEG concentrations for in vitro selection of potato plants. This research was hold at the tissue culture laboratory, the center of biotechnology development at the University of Muhammadiyah Malang starting in January 2019 until May 2019. This research used RAL with one factor which was four times repeated. The treatment used in this study was P0 = without PEG support, P1 = PEG concentration of 13%, P2 = PEG concentration of 14%, P3 = PEG concentration of 15%, P4 = PEG concentration of 16%, and P5 = concentration of 17%. The variable of this study was LD50 trial is the percentage of live explants, explants height, number of leaves, number of segments, percentage of explants sprouting, percentage of leaf explants, percentage of explants live, percentage of explants dead, chlorophyll content, stomata appearance , Fresh weight, dry weight, explant firmness, explant color. The results of this study indicate that there is a very significant effect between the treatment of PEG administration on growth variables on all observed research variables. The right PEG concentration for selecting potato explants is 13%, 14%, and 15%
