This research aims to study the effect of mango explant size and combination between active charcoal concentration and plant growth regulator of IBA (Indole Butiric Acid) on mango rooting induction through in-vitro culture. The research was conducted in Laboratory of Tissue Culture of Centre for Biotechnology Development Muhammadiyah University of Malang. A simple Completely Randomized Design (RAL) was employed in this research with treatments as follow: combined explant size (small explant < 0,5 cm), (medium explant 0,5 – 1 cm), (large explant > 1cm) and active charcoal (0,1 % concentration, 0,2 % concentration, 0,3 % concentration) as well as Indole Butiric Acid (IBA) (1 ppm concentration, 3 ppm concentration, 5 ppm concentration). The results obtained in this research showed that explant which broke up on medium explant on various concentrations of active charcoal + IBA took longer time than both small and large ones, namely ranging from 21 to 29 Days After Initiation (DAI). Such combination of small explant and combined active charcoal + IBA 0,2 % + 5 ppm showed the fastest explant breaking-up, namely 7,00 ± 5,36 DAI, while combination of active charcoal and combined active charcoal + IBA (0,1 % + 1 ppm) dan (0,1 % + 3 ppm) resulted in highest number of explant namely 7,20 ± 3,68 and 6,5 ± 3,68. not all explants growed sproutings. Combination of large explant with combined active charcoal + IBA (0,3 % + 1 ppm) revealed the fastest sprout initiation, for 17,5 ± 1,07 while such medium explant combined with active charcoal + IBA 0,3 % + 3 ppm revealed the longest sprout initiation, for 67,67 ± 2,33. number of leaf range between 2 and 5 blades, but large explant showed largest leaf on each treatments, except for active charcoal + IBA (0,1 % + 3 ppm), (0,2% + 5 ppm) and (0,3 % + 1 ppm) which formed no leaves. Large explant on various concentrations of active charcoal + IBA showed the faster rooting initiation and the latest one on medium explant.
