Abstract

The are of this research is Gram negative bacteria that can cause diarrheal disease are Escherichia coli bacterial infections or often called E. coli and some E. coli bacteria are resistant to antibiotics so it is necessary to develop new drugs that can cope with infections. Determine the secondary metabolites found in the n-hexane fraction of Limonia acidissima L fruit to obtain the zone diameter inhibition of Limonia acidissima L fraction of n-hexane fruit against the growth of Gram negative bacteria such as Escherichia coli. The methods that used to determine the antibacterial activity using the Disc Diffusion method. The n-hexane fraction of Limonia acidissima L fruit was obtained by multilevel maceration method and secondary metabolites were identified by Thin Layer Chromatography method. The test concentrations used were 100 mg / ml, 150 mg / ml and 200 mg / ml for negative controls using DMSO 1% + aquadest and chloramphenicol 30μg / disk positive control. Test and control solutions were placed on agar nutrient media which had been smeared with bacterial suspense and then incubated at 37 ℃ for 24 hours. The results of identification of secondary metabolites with Thin Layer Chromatography showed positive results on Alkaloids which gave colored stains (orange), Terpenoids (purple), Flavonoids (intensive yellow) and Anthracinone (green). The results of the antibacterial activity of Limonia acidissima L n-hexane fraction against Escherichia coli bacteria with concentrations of 100mg / ml, 150mg / ml and 200mg / ml did not provide a inhibition zone because the diameter of the clear zone was the same as the disc diameter of 6.0 ± 0 mm. so it can be concluded that the n-hexane fraction of Limonia acidissima L fruit has no biological activity as an antibacterial.
