Abstract

This research aims to analyze the characteristics of pesticide-tolerant bacteria of chili farm land in Jabung Village of Blitar Regency, as well as to determine the bacteria species that can be tolerant toward pesticides with active ingredients of acetamiprid, imidacloprid, and propineb. This is a descriptive research that emphasizes the characteristics of macroscopic, microscopic, biochemical tests, and species of pesticide-tolerant bacteria that are successful in isolation. The isolation was performed by diluting soil sample of the farm to 10-5 and it was grown by spread plate on TSA medium containing 200 ppm active pesticide ingredients. Pure isolate obtained was observed to find the characteristics of macroscopic colonies, microscopic cells, biochemical tests of cell activity and bacterial isolates identified by using MICROBACT GNB 24E (12A+12B) adapted to the book of Bergey’s Manual of Determinative Bacteriology dan Bergey’s Manual of Systematic Bacteriology. The bacteria that were able to grow in the isolation media were 13 isolates. Isolates of pesticide-tolerant bacteria included in gram-positive endospores, gram-positive non-endosporic bacteria and gram-negative bacteria. Based on the identification results, there are 8 species of pesticide tolerant bacteria, namely Pseudomonas saprofiticus, Bacillus alvei, Bacillus cereus, Bacillus sphaericus, Bacillus amyloliquefaciens, Bacillus subtilis, Bacillus firmus and Staphylococcus saprophyticus. The indicator showing that bacteria are able to tolerate pesticides in which bacterial colonies can grow on TSA medium containing 200 ppm pesticides, while intolerant bacteria cannot grow on the medium. Tolerant bacteria are able to adapt and grow on nutrient-poor medium containing pesticides, and it proves that these bacteria can utilize the pesticide active ingredients as carbon sources to support their growth. The process and results of this research can be used as a learning source of biology for 10th grade students in kingdom monera material, because it has fulfilled the six requirements for the use of learning resources, namely clarity of potential, suitability for learning objectives, clarity of target, clarity of information, clarity of exploration guidelines and clarity of acquisition.
