Abstract

Fusarium oxysporum is a member of the Tuberculariaceae family, a hypocreales order that has the potential to produce mycotoxins which are often found in animal feed or food. Fusarium oxysporum mushrooms are saprophytic and parasitic. The attack of this fungus attacks plants from various phases and at various altitudes, and results in a decrease in the production of agricultural commodities and results in losses for Indonesian farmers (Hidayat, 2017). Based on this situation, characterization of fusarium species is an important thing to learn more about. The current characterization approach can be known more deeply with the increasing knowledge in the field of molecular biology (Tjitrosoepomo, 2009). The purpose of this study was to find out the characteristics of the lowland Blus Fusarium oxysporum isolates by molecular phylogenetic analysis. The research was conducted in May to September 2018. Propagation of samples was carried out at the Agrotechnology Laboratory of the University of Muhammadiyah Malang, while the isolation of sample DNA, PCR, electrophoresis and data analysis was carried out in the Genetic and Molecular Laboratory of the Biology Department of the Faculty of Science and Technology Maulana Malik Ibrahim State Islamic University Poor. This research is a qualitative explorative descriptive study. The object of this research is Fusarium oxysporum in the lowlands of Blitar. The study was conducted to determine the molecular characterization of Fusarium oxysporum consisting of roots and lowland land of Blitar. The implementation of the study consisted of several activities including making media, preparing samples, extracting DNA, PCR-elekroforesi. The results showed that DNA isolated from Fusarium oxysporum had calculations of DNA concentrations in Blitar and Blitar root samples showing results of 8.98 and 20.72 ng / μl. The purity ratio of AB DNA is 1.07 and TB is 1.23. DNA amplification carried out through 3 PCR processes can be seen from the size of the tape produced by the band between 500bp and 750bp which is around 600bp. The genetic distance value of 0.011 against other Fusarium oxysporum DNA sequences from the NCBI GenBank database which later formed a monophyletic taxon in the evolution of the evolution of the tree shows a close kinship relationship.
