Abstract

This study aimed to assess the pattern of interaction distribution mutant endophytic bacteria transformed gfp gene in rice plant tissues in vitro. This study using a two-stage design of the phase I with Complete Random Design (CRD) is simple with Treatment ; Control, Mutant endophytic bacteria + gfp (IM-1, IM-3, IM-8, IM-24, IM-25) and Phase II with Completely Randomized Design (CRD) factorial by a first factor is Organ (O); Leaves O1 =, O2 = Stem, O3 = Roots; and second factor is mutant isolates; Control, Mutant endophytic bacteria + gfp (IM-1, IM-3, IM-8, IM-24, IM-25). Each treatment combination was repeated 3 times with dilution of 101, 102, 103, 104, 105, 106, and 107. The parameters measured were percentage of seed imbibition (%), seed germination time (dap), the number of colonies on each organ (Log cfu / ml) and the number of colonies of each isolate (cfu / ml) Results of this study on phase I concluded that the initial stages of the invasion in the pattern of interaction of bacteria on a percentage imbibition show 5 mutant endophytic bacteria + gfp isolates ranged between 4% -17%, the highest in the mutant isolates IM-3 + gfp (17%) while the lowest in the Mutant Isolates IM -8 + gfp (4%), not significantly different in seed germination time parameters, Mutants IM-1 + gfp, and IM-8 + gfp higher in stem organ, Mutant IM-24 + gfp higher in roots organs, and Mutant IM-25 + gfp higher in leaf organs. Results of this study On the phase II concluded that Mutan IM-25+gfp have average total coloni higher than another mutan with 1.643 x 109 cfu/ml.
