Abstract

Identification of cultivars with DNA fingerprinting was very important, because it was used for the protection of intellectual property, to describe the cultivars which will be released, identify the superiorities of a cultivar, knowing elders of a cultivar and early identification seedling in the nursery industry. The research purpose to identify the DNA fingerprinting 15 promising clones of sugarcane (Saccharum spp. Hybrids) and the parents based on microsatellite markers. DNA from 15 promising clones sugarcane and the parents amplified using three primer was mSSCIR5, SMC1047HA and MCSA053C10. The diversity data of electrophoresis results based on the appearance of DNA banding pattern was analyzed using analysis cluster with a hierarchical techniques that exist in the program Numerical Taxonomy and Multivariate Analysis System version 2:02 (NTSYS) were displayed in the form dendogram. Fingerprinting 15 promising clones sugarcane and the parent with 3 pairs of primer has a banding pattern that varies with the size range of 122-1975 bp. The third primer capable of displaying unique band alleged the identifier of clones, ie KB 15-8 (1110 bp), BS 21-04 (677 and 989 bp), PS881 (1251 bp), IJ76-378 (438 and 555 bp), IJ76 -375 (150 and 612 bp), IJ76-396 (236 bp), KB 14-16 (271 bp), PSJT 941 (773 bp), KB 08-2 (1975bp). The results of analysis cluster were displayed in the form dendogram shows a grouping of the 15 promising clones sugarcane and the parental based on Jaccard similarity coefficient is divided into two large groups at the similarity level of 0.23: group I and II. Group I was divided into sub-subgroups. The first group was divided into two subgroups, namely subgroups I.1 and I.2 to the similarity value of 0.33. I.1 subgroups divided into three sub-groups: (A) the similarity value of 0.45 consisting of clones BL, BS 04-01, PSJT 941, KB 18-27, PS 864, KK, PS 881, PS 862 , (B) the similarity value of 0.54 consisting of clones KB 15-16, 15-17 KB, KB 15-8, (C) the similarity value of 0.50 consisting of clones KB KB 14-16 and 14-25. I.2 sub-group consisting of (A) IJ76-378, IJ76-370, IJ76-374, IJ76-396 the similarity value of 0.59. Group II at the similarity value 0.24 consists of clones from crosses between PS 862 and IJ76-375 that was 08-15 KB, KB 08-38, BS 21-22, KB 08-2, KB 08-3, BS 21-21, BS 21-04 and IJ76-375. Clones that have the highest similarity value at the similarity value of 0.75 consisting of PS 864 clones and clones KB KB 18-27 and 08-38 and 21-22 BS.
