Abstract

Background: Escherichia coli is one of the main causes of infection. The emergence of various infections as well as the resistance caused by bacteria, encourages a continium efforts in developing herbal-based medicine that can eliminate bacteria also avoiding the occurrence of these resistance. A plant which is used as a medicinal ingredient is Limonia acidissima L. In a previous study, Limonia acidissima L. fruit pulp contained several compounds such as flavonoids, saponins, alkaloids, and tannins that have the antibacterial activity. Objective: The purpose of this research is to obtain the data of antibacterial activity from secondary metabolites found in the ethanol fraction in Limonia acidissima L. fruit against the Escherichia coli bacteria with disc diffusion method and to obtain the data of secondary metabolites contained in the fruit. Method: The method researcher used was the fractionation stages with n-hexane solvent, ethyl acetate, and ethanol in Limonia acidissima L. fruit pulp powder. Antibacterial activity test of Limonia acidissima L. pulp was done by using disc diffusion method. Results and Discussion: The results of the antibacterial activity test of ethanol fraction of Limonia acidissima L. fruit pulp by disc diffusion method, at a concentration of 5 mg/disc on average showed a diameter of 13,77 ± 1,72 mm, at a concentration of 7,5 mg/disc is 15,47 ± 2,08 mm, and in concentration of 10 mg/disc is 16,63 ± 2,11 mm and positive control of chloramphenicol 30 µg is 16,67 ± 1,05 mm. The identification of compound groups in ethanol fraction of Limonia acidissima L. fruit flesh was done by TLC (Thin Layer Chromatography) method obtained alkaloid, terpenoid, and anthraquinone compounds, while for the identification of saponin compounds was done by performing foam test and salkowski test.
