Abstract

Background: Synthesize of O-(4-trifluoromethylbenzoyl)-5-methylsalicylic acid as an salicylic acid derivate. The compound is expected to have higher analgesic activity that compare with aspirin. Objective: Produce O-(4-trifluoromethylbenzoyl)-5-methylsalicylic by reacting of 5-methylsalicylic acid and 4-trifluoromethylbenzoyl acid using acyl halide esterification method and this analgesic activity compare with aspirin in mice. Method: The synthesis uses esterification method of acyl halide, while the analgesic activity test uses stretching method (Writhing test). Result: The result obtained from synthesis is yellow crystalline, odorless with percentage 8,31%, and has melting point 156-158oC. TLC test results obtained 1 stain using 3 different eluents : n-Hexane : Ethyl Acetate (6:4), Chloroform : Methanol (7:3), Ethyl acetate : Chloroform (7:3). Identification of compound structure with the UV Spectrophotometer showed the wavelengths 205 nm, 228 nm, and 278 nm. IR spectrophotometer showed the wave number 3461,81cm-1 (-OH); 2926,81 cm-1(CH3); 1737,71 cm-1 dan 1677,68 cm-1 (C=O); 1613,79 cm-1 (C=C); 1272,72 cm-1 (COOC). The H-NMR specthrometer showed 11 H atoms. The percentage of pain inhibition for dose 25 mg/kgBB = 38,16 %, 50mg/kgBB = 53,89 %, 100mg/kgBB = 66,80%. The O-(4-trifluoromethylbenzoyl)-5-methylsalicylic acid has ED50 of 43,34 mg/kgBB while Aspirin which is the salicylic acid derivate had ED50 of 135,67 mg/kgBB. Conclusions: The O-(4-trifluoromethylbenzoyl)-5-methylsalicylic acid was synthesized and the analgesic activity higher than Aspirin.
