Abstract

The low productivity of potatoes in Indonesia is the use of seed yield no clear origin of this thing that causes seed quality decreases, the approach that is reproduction in vitro in seeds with the method mikropopagasi who can mengahasilkan seed potato pathogen-free. This research aims to know the influence of the concentration [NH4NO3] and growing regulatory substances BAP (6-Benzyl amino purin) on the growth of potato shoot varieties of Granola Swell in in vitro and get the proper concentration for optimization growth of shoots potatoes. This research was carried out in the laboratory of Biotechnology University of Muhammadiyah Malang Design used is a simple group of random design with 9 treatments and 3 replicates. The observed parameters i.e. high number of buds, shoots, the number of book, number of leaves, roots, the percentage amount of contamination, the percentage of live and the percentage of kalus. Results of the study showed that giving regulatory substances and concentrations of NH4N03 grow BAP (6-Benzyl amino purin) gives a real influence on the parameter number of the book, namely the combination of concentration 0.5 μm + 17.84 μm on (14 HST) and the number of leaves on the concentration 17.84 μm + 0.5 age (14 HST), at a concentration of 1 μm + 39.28 μm (21 HST). While the real effect on NH4N03 parameter of high number of buds and shoots occur at age 7 (HST) concentration (39.28 μm), a number of influential books for real at age 28 (HST) concentration (39.28 μm). BAP concentrations at the treatment (6-Benzyl amino purin) do not differ markedly on any parameter of the potato varieties of Granola Swell.
