Abstract

Background : O-(2-chlorobenzoyl)-5-methylsalicylic acid is a derivative of salicylic acid resulting from the synthesis of 5-methylsalicylic acid and 2-chlorobenzoyl chloride with esterification of acyl halide method. Objective : Produces an O- (2-chlorobenzoyl) -5-methylsalicylic acid compound of the reaction modification of the 5-methyl salicylate acid structure with the 2-chlorobenzoyl chloride reagent and the analgesic activity of the O- (2-chlorobenzoyl) -5-methylsalicylic acid compound and compares its activity with acetosal in mice (Mus musculus). Method : The synthesis is using esterification method of acyl halide, while the analgesic activity test using stretching method (Writhing test). Result : The result obtained from synthesis is crystalline, white, odorless with a percentage %, with a melting point 133-135oC. TLC test results obtained 1 stain using eluent as Chloroform : Methanol (7:3), Ethyl acetate : Methanol (7:3), n-Hexane : Ethyl acetate : Methanol (6:3:1). Identification of structure with the UV-Spectrophotometer showed the waveleng this 208 nm, 232 nm, dan 282 nm. IR spectrophotometer showed the wave number 3480,59 cm-1 (-OH); 2923,81 cm-1 (CH3); 1750,29 cm-1 and 1703,37 cm-1(C=O); 1610,61 cm-1 (C=C); 1205,35 cm-1 (COOC). The 1H-NMR specthrometer showed 11 H atoms. The percentage of pain inhibition for dose 25 mg/kgBW = 29.25 %, 50mg/kgBW = 51,77%, 100mg/kgBW = 60,97%. The O-(2-chlorobenzoyl)-5-methylsalicylic acid has ED50 of 56,19 mg/kgBW while acetocal which is the salicylic acid derivate had ED50 of 135,67 mg/kgBW. Conclusions : The O-(2-chlorobenzoyl)-5-methylsalicylic acid was synthesized and higher activity than acetocal.
