Abstract

Backgrounds : Breast cancer is one of the most common types of cancer that is commonly experienced by both women and men in Indonesia. Breast cancer is a malignant tumor that is formed from breast cells that grow and develop abnormally and uncontrollably that can spread to the tissues near the breast or other parts of the body due to metastatic processes that occur. Cancer therapy in general has a relatively greater side-effect compared to the benefits obtained, so treatment by using natural ingredients is being intensively conducted. Sawo manila (Manilkara zapota) is a plant that has the potential to have anticancer activity. The research aims to determine the cytotoxic effect of sawo manila seeds (Manilkara zapota) on breast cancer cells by MTT-Assay method and to identify the classes of chemical compounds contained in the ethanol extract of sawo manila seeds using Thin Layer Chromatography Method : The cytotoxicity test of ethanol extract sawo manila seeds using 4 concentrations, ie 1000 μg / ml; 900 μg / ml; 400 μg / ml and 300 μg / ml with a cell density of 104 cells per well. Doxorubicin as a positive control uses 5 concentrations, ie 100 µg/ml ; 75 µg/ml ; 50 µg/ml ; 25 µg/ml and 12,5 µg/ml. Identification of classes of chemical compounds contained in the ethanol extract of sawo manila seeds using Thin Layer Chromatography test with Kiesel GF 254 as stationary phase and n-hexane – Ethyl Acetate with ratio (9: 1) as a mobile phase. Stain markers used in identification were Dragendorff, FeCl3, 10% Sulfuric Acid, Anacaldehyde Sulfate Acid and KOH 10% in methanol. Result and Conclusion : Result of cytotoxic test showed that ethanol extract of sawo manila seeds (Manilkara zapota) was not active in giving anticancer activity with IC50 value 193,177 μg / ml. TLC results indicate that ethanol extract of sawo manila seeds have alkaloid and terpenoid.
