Abstract

Background: The aim of this research is to develop a new analgesic drugs, synthesize of Acid O-(4-methylbenzoyl)-5-methylsalicylate as an acid salicyl derivate. The compound is expected to have higher analgesic activity than aspirin. Objective: Produce a new compound of salicylic acid derivate, Acid O-(4-methylbenzoyl)-5-methylsalicylate by reacting 5-methyl salicylic acid and 4-methylbenzoyl chloride with esterification method of acyl halide. Analgesic activity will be tested and then compare with aspirin. Method: The synthesis is using esterification method of acyl halide, while the analgesic activity test using stretching method (Writhing test). Result: The result obtained from synthesis is crystalline powder, yellowish, odorless with a yield percentage of 27,61% with a melting point of 143-145 oC. TLC test results obtained 1 stain using eluent: Ethyl acetate: Chloroform (7: 3), Ethyl acetate : Methanol (7: 3), Aceton : Methanol (6: 4). Identification of structures with the UV Spectrophotometer showed the wavelengths 204 nm and 238 nm. IR spectrophotometer showed the wave number 3463,63 cm-1 (-OH); 2925,60 cm-1 (CH3); 1692,43 cm-1 and 1746,44 cm-1 (C = O); 1610,57 cm-1 (C = C); 1265.45 cm-1 (COOC). The 1H-NMR specthrometer showed 14 H atoms. The percentages of pain inhibition for dose 25mg/ kgBW = 37.33%, 50mg/kgBW = 53,72%, 100mg/kgBW = 69.89%. The Acid O-(4-methylbenzoyl)-5-methylsalicylate had ED50 of 45,26 mg/kgBB while the Aspirin which is the salicylic acid derivative had ED50 of 135,68 mg/kgBB. Conclusions: Acid O-(4-methylbenzoyl)-5-methylsalicylate was synthesized and had higher activity than aspirin.
