Abstract

Apples contain substances that can improve the body health. Some phytochemical compounds present in apples that act as antioxidants are flavonoids, tocopherols, phenolic, coumarins, cinnamic acid derivatives, and polyfunctional organic acids. In addition, apples contain beta-carotene which serves as a provitamin A to prevent free radical attack One way of producing secondary metabolites is to use precursorsin in vitr culture. The purpose of this research was to know the response of combination of sucrose and phenylalanine on different MS medium to quercetin production through apple callus culture. This research used Factorial Randomized Block Design. The treatments were apple varieties (V1 = Manalagi V2 = Granny Smith) and precursors with treatment M0 (no additional precursors), M1 (phenylalanine 500 mg/l) , M2 (sucrose 10 g/l), M3 (sucrose 15 g/l and phenylalanine 200 mg/l), M4 (sucrose 10 g/l and phenylalanine 400 mg/l), and M5 (sucrose 5 g/l + phenylalanine 600 mg/l). The data obtained was calculated by the analysis of the variance then analyze through honestly significance difference (HSD) test on 5% level. The results showed that callus of V2M3 gave the best result on parameter length, width, surface area, fresh weight, and dry weight of callus. The higher total flavonoid (quercetin equivalent) obtained by V2M3 ie 147.57 mg/g dw, antioxidant activity of callus with concentration of 50% free radical resistant DPPH (1,1-diphenyl -2-picrythhydra-zil) was lowest on V2M3 ie 1486.76 ppm callus extract; and V1M2 ie 1494.70 ppm callus extract.
