Abstract

Plasmid is a circular and small-sized extracromosomal DNA (1 - 200 kb). Most procaryotic organisms, in general bacteria, have plasmids. Plasmids are capable of autonomous replication, independent of chromosome replication. However, there are several plasmids whose replication follows the replication of chromosomal DNA. Bradyrhizobium is one of the bacteria that has the ability to provide nutrients for plants. If the bacterium is symbiotic with legume (nuts), it will infect the roots of the plant and form the root nodule inside. This research was conducted at the Laboratory of Biotechnology University of Muhammadiyah Malang which lasted from April to August 2016. This research aims to obtain Bradyrhizobiumjaponicum plasmid that is resistant to 5 types of contact pesticides. In this research use 6 treatment that is control, active substance pesticide Beta Silfutrin g/l,active ingredients Prefenofos 500 g/l,active ingredient Deltamethrin 25g/l,ctive ingredient Diafentiuron 500 g/l, danctive ingredient Alfametrin 15 g/l. The results showed that there was a change of color of the solution on the media used for plasmid isolation. Which initially the solution looks clear after mixed with the treatment and in the shaker turns the color becomes visibly turbid. Based on electrophoresis results from plasmid DNA isolation showed the presence of two parts of the plasmid DNA bands that appeared. At the top of the DNA band has a size of 180 pb, while the bottom has a size on the active ingredient treatment Deltamethrin25 g/l (P3)with a size between 30-40 bp and other treatments having the same plasmid DNA size ranging from 20-30 bp.
