Abstract

Mango cultivars like Arumanis 143 superior Indonesia experienced the inability to compete in international markets because of the color is less attractive Arumanis 143 is bluish green, while consumers prefer the mango fruit that has yellow, orange, red fruit on the skin's appearance.One effort to improve the attractiveness of mango Arumanis 143 performed by crossing these cultivars with mango cultivars which have the skin color of yellow, orange and red. The ideal character of mangoes for export or for fresh consumption is the color of the skin and meat (red - yellow - orange), fruit flesh thickness with thin beans, taste, aroma, texture and fiber like Arumanis 143. This research was conducted at the Laboratory of Agricultural Cultivation Department Biotektonologi Fak. Agriculture UB using three crossbred varieties of mango plant which is Arumanis 143, Haden and Irwin and red clones Haden and Irwin numbering 24 who came from Garden Collection Germplasm Research Institute for Fruit CukurGondang Pasuruan. PCR amplification process - SSR done using a type of primary SCARIS 18. This research was conducted with four stages ie first stage of sampling, DNA isolation, amplification of DNA with PCR reaction, gel electrophoresis agrosa, further results in the form of photographic images of DNA using UV light. Results showed that 18 SCAR markers are DNA markers can be used to characterize differences in skin color of fruit on the three elders of the mango crop is Arumanis 143, Haden and Irwin. SCAR markers 18 which is a marker of DNA can be used to identify genes that cause differences in skin color of fruit on the results of three crosses between parental plants namely mango Arumanis 143, Haden and Irwin. Results cruciferous Arumanis 143 x Haden got crosses of accession of the F1-21, F1-46, F1-86, F1-88,, Haden x Arumanis 143 accessions was found that crosses of the F1-30, F1-94, which shows the pattern of DNA bands identified a type of fruit peel red color using simple techniques and repeat sequence primers SCAR 18. Accession hybrid crosses between Arumanis 143 x Haden namely F1-15, F1-27, F1-26, accession to Haden x hybrid crosses between the F1 Arumanis 143-2001, F1-02, F1-42, accession Arumanis 143 x hybrid crosses between the F1-Irwin 22, F1-35, F1-69, accession Irwin x Arumanis hybrid crosses between the F1-08 143, F1-09, F1-10, F1-11, F1-14, F1-36, F1-29, F1-31, F1-43 shows the length and number of the same DNA banding pattern with the pattern of parental Arumanis 143 tape so it does not identify the type of characterization of the red color to the size of their DNA banding pattern - each is 200 bp, 400 bp.
