Abstract

This research conduced to finds the effectivity of preservative chlorobutanol 0.35% b/v in the preparation injection difenhidramine hydrochloride multiple dose after the seal packaging openend and injected first time. The effectivity of preservative could be seen by doing the sterility test by inoculating sample aseptically into medium named thioglikolat and kassamino. Sample diluted before to remove the antimicroba activity. The dilution comparison is 1:2 for thioglikolat medium and kassamino medium by replicating it for 3 times. The tes were done 5 times in 28 days storage period in 1st, 7st, 14st, 21st, and 28th day. Sample then incubated in the temperature 32,5°C ± 2,5°C for no less than 14 days for thioglikolat medium, and kassamino medium at 22,5°C ± 2,5°C for no less than 14 days. As the sterility result indicator we made positive that show the presence of microorganism growth and we made negative control that show the preparation were sterile. As the positive control we implanted bacteria Pseudomonas aeruginosa to thioglikolat medium and fungi Candida albicans to kassamino medium. From the research we got the result that effectivity of preserrvative chlorobutanol with the consentration 0.35% blv had fullfil the requirements of USP that cold sustain the preparation injection difenhidramine hydrochloride multiple dose still sterile by five times sample testing in 28 days storage period after seal packaging opened and injected for first time.
