Each year the demand for energy continues to rise, but world oil production continued to decline after 2020 (Mittelbach, 2006). Indonesian fuel consumption in 2004 reached 60 billion liters/year, of which 22 billion diesel, kerosene 12 billion, 20 billion premium and other fuel oils 6 billion (Hamdi, 2005). Therefore we need a renewable energy source instead of petroleum which is currently the main energy source, it is included in energy diversification. Biodiesel is a vegetable oil obtained from plants have many advantages than other energy sources (Subandi, 2009). Analyzing and evaluating transgenic plants jatropha (Jatropha curcas L.), which has been transformed with the plasmid vector containing pCambia 1304 gusA gene, through molecular analysis and analysis. The research was conducted at the Laboratory of Molecular plants, Pusbang Biotechnology University of Muhammadiyah Malang. The study was conducted from October 2011 to April 2012. The materials used include: young leaves of Jatropha plants from 30 numbers (Table 1). While the chemicals used were: extraction buffer, lysis buffer, CI 24:1 (Chloroform: isoamyl alcohol), absolute ethanol, ethanol 70%, TE buffer, 1x TBE, agarose gel, 6x loading dye, EtBr (ethidium bromide), dH2O, 100 bp lambda DNA, nuclease free water, PCR mix, and plasmid pCambia 1304 From the research that has been done, 29 of 30 plants (96.67%) showed positive transformation of gusA gene in the plant Jatropha with molecular testing, the test root mapaun leaves histochemistry. Molecular test (PCR) showed that the gusA gene has been well integrated into the genome of Jatropha plant is shown by 22 of 30 plants (73.33%). Histochemical test leaves showed gusA gene interaction with X-gluc giving rise to blue patches on the leaf tissue in 24 of 30 plants (80%). Histochemistry root test showed gusA gene interaction with X-gluc giving rise to patches of blue in the root tissues of 8 of 30 plants (26.67%).
