Abstract

Orchids aclimatitation medium that usually used are charcoals, sphagnum, edible fern, husk or wood skin, where the available of nutrition in medium were limited. These limited of aclimatitation medium nutrition was being the idea of this research to modifying the aclimatitation medium to be better, where the medium nutrition could be available in reasonable amount. These modified medium in this research were using the culture in vitro medium which added to the aclimatitation medium. Nevertheless, in this application the medium contamination is the problem. So, to solving the contamination antimicroorganism increment was necessary, one of the antimicroorganism that could be used is Methyl Paraben (C8H8O3). The purpose of this research is to examine the effectiveness of the Methyl Paraben (C8H8O3) which combined with pH level to reduce the aclimatitation medium contamination which added by in vitro medium and how the effect to the orchids Blc. Chialin Beauty ‘Golden Eyes’ growth. The research was held in Agroorchids Nursery green house of the University of Muhammadiyah Malang, since October 26th 2011 to January 18th 2012. Using factorial Randomized Block Design, with two treatment factor which each of the treatment consist by five level, and each level repeated three. The value of this research indicated that the concentration and pH ex vitro medium were reduced the contamination in all of the treatment combination. Nevertheless, this treatment could not increased the plant growth which include plant tall, number of leaves, number of roots, roots long and fresh weight, except in leaves wide at age 5th, 8-12th weeks after planting.
