Abstract

Biological nitrogen fixation (FNB) is a process of symbiosis between legume plants with bacteria. The result is the availability of nitrogen required by the host plant and used also by non-legume plants growing around the symbiotic environment. One of the bacteria that play a role in the belay nitrogen in legumes root bacteria Bradyrhizobium japonicum is a symbiosis with soybean plants. Bradyrhizobium japonicum bacteria are inhabitants of soil microbes that infect the roots of legume plants to form root nodules. The symbiosis between legume plants and bacteria is mutually beneficial to both parties. Bradyrhizobium japonicum obtain nutrients (fotosintat) that is rich in energy from the plant host while the host plants obtain nitrogen compounds from these bacteria to continue to thrive. The purpose of this research is Bradyrhizobium japonicum to obtain isolates that are resistant to the antibiotics chloramphenicol and tetracycline. The study was conducted at the Laboratory of Biotechnology Muhammadiyah University of Malang. The research began in the month of April 2011 until January 2012. Plasmid isolation method used is the method of alkaline lysis with SDS in the adaptation of Sambrook and Russell (2001). The results showed that of the 9 isolates tested, 4 isolates of Bradyrhizobium japonicum were resistant to chloramphenicol 5 mg / ml, which is a row of which is the most resistant isolates 1, 3 isolates, isolates 9, and 8 isolates. Meanwhile, the use of the 9 isolates, none of which were resistant to tetracycline 5 mg / ml.
