Abstract

Pinneapple (Ananas comosus) is a plant in Parana river bank area. Pinneapple has potential as export plant. The largest export volume for culture commodity consisted of processed pinneapple was 49,32% of all total Indonesia’s culture plant export at 2004. Pinneapple has bromelin enzyme (protease enzyme which could hydrolized protein, protease, or peptide) Bromelin could be extracted directly or produced in in vitro way. The benefit of in vitro production process was didn’t depend on season, time, reduce the land-use, consistent quality and produced as the market needed. The research aimed to find out the different influence of NAA and BAP at MS media to pinneapple shoot and kalus to detect Bromelin enzyme at pinneapple explant. The research was held in Bioternologi Laboratorium University of Muhammadiyah Malang in August 2008 to January 2009. The research used simple complete random design. The research was done 3 times with interval once in 10 days, with observation parameters: explant length, root explant percentage, kalus explant percentage, shoot explant percentage, color, wet weight, dry weight, enzyme detection. According to the research, there found an interaction between BAP and NAA in the growth control element of BAP and NAA to the brown explant color percentage 20 and 30 days age after plantation, white color explant percentage 20 and 30 days age after plantation, wet weight 10 and 20 days age after plantation, and dry weight 30 days after plantation. BAP 1 ppm + NAA 7,5 ppm was better since it force the kalus and shoot growth, green explant color was higher with increasing wet and dry weight. Enzyme activities was influenced by BAP growth control element in 1 ppm concentrate. BAP 1 ppm + NAA 7,5 ppm and BAP 1 ppm + NAA 15 ppm treatment were the best treatment in enzyme activity, the treatment increased enzyme activities along the growth.
